That the hydrolysis of β, β-trehalose by the purified acid-stable and less acid-stable saccharogenic amylase preparations of Aspergillus awamori var. fumeus-1-B42 (hereafter to be abbreviated as AF-1-B42) is effected by these saccharogenic amylases and not by contaminated enzymes, was made sure by the following results. (1) Identical pH-activity curves were obtained for saccharogenic amylase and β, β-trehalose hydrolyzing activities with the purified amylase preparations; (2) The inactivation of saccharogenic amylase activity at various pH paralleled with that of β, β-trehalose hydrolyzing activity; (3) The inactivation of saccharogenic amylase activity at elevated temperature also paralleled with that of β, β-trehalose hydrolyzing activity; (4) The inhibitors affecting to saccharogenic amylase activity also inhibited β, β-trehalose hydrolyzing activity at the same extent.
