The effect of mechanical damage on the production of maridomycin by Streptomyces hygroscopicus AV-901 was studied usng a shaking flask containing glass beads (B-flask) to improve the fermentation characteristics. The leakage rate of nucleic acids related compounds from the microbial cells was measured in a 200-ml Erlenmeyer flask containing several kinds of beads. An empirical equation for the leakage rate with the B-flask is presented. The fermentation characteristic values in the B-flask containing a glass bead of 4.9 mm diameter (Bl-flask) agreed with those in the 200-l fermentor under the standard conditions. Using the Bl-flsk, the effects of inoculum ratio, temperature and the concrntration of inorganic phopsphate in the medium on the maridomycin production were kinetically analysed and the optimum conditions in the 200-l fermentor were estimated.
