Exogenous Pyrophosphate and Polyphosphates Used as Energy Sources for Anaerobic Removal of Organic Matter by Activated Sludge

Intracellular polyphosphate (polyP)-deficient activated sludge in a laboratory-scale batch anaerobic-aerobic system was produced under conditions of orthophosphate (P_i) starvation. This sludge showed quite slow removal of dissolved organic carbon (DOC) during the anaerobic phase. The addition of inorganic pyrophosphate (PP_i) to the P_i-starved system during the anaerobic period stimulated DOC removal significantly. Externally added short-chain-length polyPs exerted such stimulatory effects as well. On the other hand, these effects were not clearly seen in a normal P_i-loaded anaerobic-aerobic system. The stimulatory effects of PP_i and polyPs were sbolished by the addition of an excess of P_i Enzyme assay experiments showed that cellfree extracts of anaerobic-aerobic sludge had pyrosphatase and polyphosphatase activities constitutively, while the activities of intact sludge itself were induced by P_i starvation. The results obtained provide evidence that P_i-starved activated sludge can use exogenous PP_i and polyPs as energy sources for DOC uptake under anaerobic conditions.
