Ammonium Ion-Depressed Fermentation of Tylosin by the Use of a Natural Zeolite and Its Significance in the Study of Biosynthetic Regulation of the Antibiotic

When a natural zeolite was added to the growth medium, tylosin production by Streptomyces fradiae increased three-fold without substantial changes in growth and pH levels. The ammonium ion concentration in the supplemented medium remained at lower levels than in the control culture. The addition of ammonium salts resulted in a decline of tylosin production. It is thus suggested that tylosin biosynthesis is subject to negative regulation by ammonium ion, but is enhanced in the presence of a natural zeolite which created ammonium ion-depressed fermentation conditions. The production of protylonolide, an intermediate of tylosin biosythesis, was also elevated by the zeolite. The results suggest than the inhibition of tylosin biosynthesis by ammonium ions occurs at step(s) involving protylonolide biosynthesis.
