A Quantum Chemical Study extended to the Sulfamyl Group of Thiazide Diuretics

HUCKEL's LCAO-MO method was applied to the sulfamyl parts of thiazide diuretics. Resonance integrals of sulfur-oxygen bond, sulfur-nitrogen bond and sulfur-carbon bond were assumed as 0.56, 0.48 and 0.60 respectively on the basis of the electronic structure of sulfur, that is, dsp^2 + (3p)^2 hybridazation. K values of the lowest vacant energy level of thiazide diuretics were less than those of DPN. Thiazide diuretics seemed to have a possibility of interfering with the biological electron transfer system. The sulfamyl group attached to the benzene ring of thiazide diuretics, which was indispensable to their diuretic action, might be related to the quantum chemical property of a nucleophilic reaction. A strong charge confrontation in both sulfur-oxygen bonds was pointed out. Further iteration is now under calculation.
