Effects of Trapidil and Suramin on Growtjh Factor-induced Calcium Response and Tyrosine Phosphorylation in Human Glioma Cells

trapidilやsuraminはPDGFやEGFなどの増殖因子によるglioma細胞の増殖を抑制するが、その機序は充分には解明されていない。本研究では、T98G細胞のPDGF及びEGFの増殖シグナルに対する両薬剤の効果を調べた。trapidilとsuraminは共に、PDGFによる紬胞内のCa^<2+>濃度の増加とそのreceptorのチロシンリン酸化を抑制した。また、両者は、EGFによる細胞内のCa^<2+>濃度の増加を抑制したが、そのreceptorのチロシンリン酸化は部分的に抑制されるにとどまった。以上より両薬剤は、増殖因子のreceptorへの拮抗作用以外に、PDGF, EGFに共通の電位非依存性Ca^<2+>チャンネルを抑制するなど、複数の経路から増殖抑制作用を示すと考えられた。
