STUDIES ON THE RADIAL TRANSPORT AND METABOLISM OF PHOSPHATE IN CORN ROOTS USING THE ^<32>P AND ^<33>P DOUBLE-LABELING METHOD

For studies on the metabolism and transport of phosphorous compounds in plants, radioactive phosphorus is widely used. Many previous experiments have been done employing ^<32>P in combination with paper chromatography (1), which is useful for separating trace amounts of phosphorous compounds (2). In those studies, however, the specific activity of the individual compound was rarely given, because of the difficulty in analyzing trace amounts of phosphorus, although it is a very important index in the study of phosphorus metabolism. A new index, the "Relative Specific Activity" (RSA : ^lt:32>P/^<33>P), was introduced instead. After continuous feeding of ^<33>P for a long period, corn seeding were supplemented with ^<32>P for a short time, and the RSA (^<23>P dpm/^<33>P), was measured. Here, a simultaneous determination method for ^<32>P and ^<33>P counts using a liquid scintillation counter is described. Also, results of application of this new index to plants combined with a root separation method into cortex and stele are given.
