Intestinal Absorption of Lactoferrin Molecules, an Immunohistochemical Study in Rats

Lactoferrin is a component of milk and other secretions in mammals, and has been known as a protein associated with iron transport and antibacterial action. Recently, lactoferrin from bovine milk, which is being used as a health food, has been reported to react directly with hepatitis C virus, reducing the virus load and improving the liver function of patients with hepatitis C. In general, proteins are considered to be digested down to amino acids and oligopeptides before they are absorbed by the intestinal tract. Studies of the absorption of lactoferrin from the intestinal tract so far have been conducted mainly from the biochemical viewpoint using cultured cells, and in vivo morphological studies are extremely rare. In the present study, we used immunohistochemical studies to examine whether lactoferrin molecules with preserved antigenicity are absobed from the rat intestine. Light and electon microscopic findings showed that a part of the lactoferrin molecules (a basic protein) was absorbed undigested and with antigenicity preserved, from the intestinal tract via pinocytosis. Lactoferrin molecules taken up into the cell passed through the cell, was secreted into the intracellular space between absoprtive epithelial cells and reached the subepithelial cell basement membrane. The lactoferrin molecules then passed through the basement membrane of the blood vessel, reaching the blood stream and was finally absorbed into the body.
