Sensory and Motor Neurons Expressing mRNAs for Neurotrophin Receptors in the Rat, with Special Reference to Changes in Biosynthesis Following Sciatic Nerve Transection

In order to understand the roles of neurotrophins in sensory and motor neurons under normal and pathological conditions, we examined the cell-size of neurons expressing trk family and low-affinity neurotrophin receptor (LANR) mRNAs, co-localization of these mRNAs, and their biosynthetic changes following peripheral nerve transaction by means of in situ hybridization. The trkA, trkB, and trkC mRNAs were expressed in approx. 35-40%, 5%, and 15-20% of rat dorsal root ganglion neurons (L5), respectively, and the cell-size distributions were different in each cell group (trkA-expressing cells : small to medium, trkB-expressing cells : medium, trkC-expressing cells ; medium to large). The trkB mRNA-expressing neurons lacked trkA and trkC mRNAs, while a few sensory neurons expressed both mRNAs. Each member of the trk family was simultaneously coexoressed with LANR. The incidences of sensory neurons expressing trk family and LANR mRNAs were decreased in a similar fashion one week after sciatic nerve transection. The trkB mRNA was expressed in non-neuronal cells surrounding DRG neurons in the present study, and levels of this mRNA were also reduced by the same treatment. 0n the other hand, trk family gene expression was unaffected in motoneurons after the same treatment, although the level of LANR mRNA was markedly increased. The present study demonstrated the differential regulation of trk family and LANR genes in sensory and motor neurons as well as in normal and pathological states, suggesting the functional multiplicity of neurotrophins on various cells under various conditions.
