Non-obese diabetic (NOD) mice, as an established animal model of IDDM, shows various immunological abnormalities including the generation of autoantibodies and abnormal production of cytokines. In the present study, the in vitro production of interleukin 6 (IL-6) and interleukin 1 (IL-1) by spleen cells in female NOD mice was investigated in order to determine how the cytokines are related to the development of diabetes in NOD mice. The production of IL-6 and IL-1 by spleen cells was measured in the culture supernatants after the incubation of spleen cells with 25μg/ml of lipopolysaccharide (LPS) for 72 hours. When aging effects were examined, the LPS-induced IL-6 production in NOD mice showed an increase at 4 and 16-18 weeks of age, whereas this increase was not observed in the control Institute of Cancer Research (ICR) mice. IL-6 production was significantly higher in NOD mice with spontaneous diabetes than in age-matched NOD mice without diabetes. Similar results were obtained regarding IL-1 production in NOD mice ; IL-1 production by spleen cells increased at 2-4 and 12-18 weeks of age, and diabetic mice showed significantly higher production of IL-1 than non-diabetic mice. In ICR mice, there was no increase of IL-1 production at 12-18 weeks of age. When NOD mice (15 weeks of age) were treated with cyclophosphamide (CY), an accelerator of diabetes in NOD mice, LPS-induced IL-6 and IL-1 production in the spleen cells of NOD mice was significantly increased after both 1 and 2 weeks in CY-treated mice but not in saline-treated mice.
