Inflammatory bowel diseases (IBDs) are chronic and refractory disorders of unknown etiology. Their entity is generally regarded as an uncontrollably increased non-specific inflammatory reaction in the intestinal tract. Among inflammatory mediators, cytokines have particularly important pathophysiological roles in IBDs and tissue concentrations of inflammatory cytokines have been correlated with the disease activity. It has been reported that the production of inflammatory cytokines in monocytes, macrophages, and other cells is inhibited through activation of the peroxisome proliferator activated receptor-gamma (PPARγ). In an attempt to clarify the effect of PPARγ on inflammatory reactions in the colon, we investigated how the activation of PPARγ influenced the production of interleukin (IL)-8 and monocyte chemotactic protein (MCP)-1 in Caco-2 cells, a cell line established from human colorectal carcinoma. IL-1β-stimulated the production of IL-8 and MCP-1 by Caco-2 cells. IL-8 production in IL-1β-stimulated Caco-2 cells was significantly suppressed by troglitazone, a PPAR-r ligand, in a dose-dependent manner. The suppressive effect of troglitazone was also observed on IL-1β-stimulated MCP-1 production in Caco-2 cells. These results suggest possible clinical effectiveness of troglitazone in the treatment of IBDs.
