Background. Adipocytes produce the physiological effectors, adipocytokines, and also play an important role in nitric oxide (NO) production in diabetes mellitus. However the role of NO in adipocytokine production is unclear. We examined whether NO had the effects on insulin resistance and the production of leptin and tumor necrosis factor-α (TNF-α). Methods. We administered Nω-nitro-L-arginine-methyl-ester (L-NAME, 10mg/100ml) solution, NO synthase inhibitor, to Otsuka Long-Evans Tokushima Fatty (OLETF) rats, a type 2 diabetic model, and to Long-Evans Tokushima Otsuka (LETO) rats, non-diabetic control. Blood levels of leptin and TNF-α were measured in the course of the experiment. We also investigated the effects of NO suppression on adipocytokines production in 3T3-L1 adipocytes, a murine adipocyte cell line. Results. L-NAME administration significantly elevated blood pressure at day 28 in the LETO rats and at day 14 and 28 in the OLETF rats, which suggested that L-NAME was effective for NO reduction. Blood levels of leptin and TNF-α in the L-NAME-treated rats were higher than those in no-treated rats. L-NAME induced insulin resistance at day 28 in the LETO rats and enhanced that in the OLETF rats. Leptin concentrations in the culture supernatant of 3T3-L1 adipocytes were increased by L-NAME in a dose-dependent manner (1-100μ mol/l). Conclusions. L-NAME enhanced production of adipocytokines in in vivo and in vitro experiment. These results provide a novel clue for elucidating the regulatory mechanism of adipocytokine production and the pathogenesis of type 2 diabetes mellitus.
