Induction of Rat Cystatin S in Submandibular Glands by Oral Administration of Capsaicin

We investigated the effects of dietary capsaicin (a pungent ingredient of hot red pepper) on the rat submandibular gland and its secretions. Several groups of animals were offered either control diets or diets containing capsaicin (from 0.0001 to 1.0%) for seven days. The rats could take the diets (under 0.1% capsaicin). However, the concentrations suppressed food consumption for two days. The weight of the submandibular glands and total saliva protein in capsaicin diet groups increased in a dose-dependent fashion, and a new protein appeared in the submandibular saliva. Immunochemical and electrophoretic properties of the protein was identical to those of isoproterenol-treatment and papain oral administration. The protein increase in a dose-dependent fashion. These results suggest that dietary capsaicin induces cystatin S in submandibular saliva. The protein likely facilitates ingestion of diets containing the irritating substance. Irritating dietary substance such as cysteine proteinase, tannic acid and capsaicin in plants alter the morphology of salivary glands and their secretions. Such alterations can be one line of protection from toxic or irritating substances in food.
