胚性幹細胞(ES 細胞)と iPS 細胞は再生医療において重要な役を果たすと期待される。糖鎖がエピトープとなる細胞表面マーカーは,これらの多能性細胞を他の細胞と区別したり,分化の過程を追跡するのに有効である。マウスの ES 細胞のマーカーとして最も確立されているのは SSEA-1 である。SSEA-1 のエピトープは Lewis X,すなわち Galβ1-4(Fucα1-3)GlcNAc である。ヒトの ES 細胞のマーカーは SSEA-3, SSEA-4, TRA-1-60 そして TRA-1-81 である。SSEA-3 と SSEA-4 はグロボシリーズの糖脂質上の抗原であり,TRA-1-60 と TRA-1-81 はケラタン硫酸に関連した抗原である。iPS 細胞は同一種の ES 細胞と同じ糖鎖マーカーを発現している。始原生殖細胞由来の多能性細胞である EG 細胞はヒト,マウスいずれでも SSEA-1 を発現している。初期胚細胞の,分岐した高分子量ポリ-N-アセチルラクトサミンであるエンブリオグリカンはマウスの ES 細胞そしてマウスおよびヒトの胚性腫瘍(EC)細胞に発現している。エンブリオグリカンは SSEA-1 など,多くの糖鎖マーカーを担っている。Iβ1,6-N-アセチルグルコサミン転移酵素(IGnT)を欠損した ES 細胞ではエンブリオグリカン合成能が低下し,4C9 抗原が消失するとともに,ラミニンでコートしたプレートへの接着能が低下した。この結果はエンブリオグリカンがインテグリン活性を増強する可能性を示す。
