Hypocholesterolemic effects of naturally occurring estrogens, estradiol and estrone, were compared with those of stilben-type estrogens, hexestrol and diethylstilbesterol, in castrated male Wistar rats weighing 120 to 140g. It was demonstrated that there was a difference in response patterns according to the two types of estrogens, when they were administered subcutaneously once daily for 20 or 60 days. Plasma cholesterol level was markedly reduced by the consecutive treatment of estrogen, 300,,2g per day for 3 days. This initial fall of plasma cholesterol was induced by all estrogens tested so far, as previously reported (Uchida and Miyake, 1961). In the rats receiving steroidal estrogen, the plasma cholesterol restored to the normal level on the 20th day of the treatment, and significantly increased on the 60th day. The ratio of cholesterol against phospholipid in plasma was reduced by all estrogens regardless of their structures or of the duration of treatment. No remarkable difference was obtained according to the compounds in their effects on seminal vesicles, ventral prostate, thymus, adrenals, epididymal fat, perirenal fat and body weight. Testosterone propionate and progesterone caused no change in plasma lipid levels. Norethynodrel (17α-Ethynyl-l7-hydroxy-5 (10) -estren-3-one) produced the decrease of plasma cholesterol with similar pattern to that caused by the stilben-type of estrogen. Cortisol acetate produced the increase of both cholesterol and phospholipid level in the similar experiment. Discrepancies appeared in the previous reports of other workers on the estrogen effect on rat plasma cholesterol were discussed and somewhat resolved with our results.
