Calvaria bone cells (bone cell) which were isolated from new born rat calvaria were investigated histologically and could induce the calcification and new bone formation in in vitro culture with β-glycerophosphate. When effects of pO2 on the proliferation of in vitro calvaria bone cells were investigated, the amount of fluorometric DNA, alkaline phosphatase, and osteocalcin which were produced by bone cells were the most in 13% O2 tension.Bone cells had a strong potential of new bone formation in vitro and had an intensive effect of pO2. PO2 may have the important effect on osteoblasts in bone formation.
