Effects of cortisol on the biosynthesis of chondroitin sulfate in chick embryo femora were studied in tissue culture. The femora from 9-day chick embryos were cultivated by the roller-tube method in various natural and synthetic media containing <SUP>35</SUP>S-sulfate. There was a wide variation in the effects of cortisol on radiosulfate incorporation into the bones according to the type of culture medium; cortisol at a concentration of 10<SUP>-5</SUP> M markedly suppressed the radiosulfate uptake of the bones growing in synthetic media, whereas the steroid showed rather stimulatory effect on the bones cultivated in certain natural media containing chick embryo extract and horse serum. The effect of cortisol was also found to vary with the concentration of the steroid, duration of culture, and developmental stage of the bones, but not always with the elongation and dry weight increase in parallel relationship.
