THE MEASUREMENT OF TISSUE ACID MUCOPOLYSACCHARIDES BY USE OF CETYLPYRIDINIUM CHLORIDE

A method was described which permits the separation of mixture of acid mucopolysaccharides and the quantitative determination of each one in tissues. It consisted of making a precipitation of acid mucopolysaccharide with cetylpyridinium chloride, eluting each one with step-by-step increasing sodium chloride or pottasium chloride solution, and then determining fhe uronic acid content in each eluent by the carbazole method of Dische.By use of this technic, the measurements of acid mucopolysaccharides in such tissues as fibrosed liver, croton oil abscess, sponge biopsy connective tissue and newly formed connective tissue around the implanted glass plate, were carried out.With the extent of hepatic fibrosis of rats, acid mucopolysaccharide value in the liver, especially hyaluronic acid and chondroitinsulfuric acid, increased considerably and its increase preceded hydroxyproline rise. And in the experimentally induced wound tissues of rats, too, similar increase of acid mucopolysaccharide was observed.
