Effect of thyrotropin releasing hormone (TRH) on electroencephalographic findings and brain monoamine metabolism in acute hepatic failure.

The effect of thyrotropin releasing hormone (TRH) on hepatic encephalopathy was investigated in rabbit galactosamine hepatitis. After tne development of overt encephalopathy, TRH was continuously administered for 2 hours (0.4mg/kg/2hr). Improvement of electroencephalographic findings such as the disappearance of slow wave activities were observed in 5 of 9 cases (TRH responders). After the administration of galactosamine, noradrenaline and dopamine concentrations in the mid-brain and pons/medulla markedly decreased both in those with and without the TRH treatment. Howevor, the concentrations of dopamine metabolite HVA and serotonin metabolite 5-HIAA were significantly higher in those treated by TRH. The TRH responders had significantly higher mid-brain noradrenaline and HVA concentrations than did the TRH nonresponders. It is suggested that the effect of TRH may be attributable to the stimulation of the dopaminergic neuron system and closely related to the noradrenergic neuron system.
