Induction of 2-5 ASmRNA in peripheral blood mononuclear cells of patients with chronic viral hepatitis during interferon treatment.

We measured (2'-5') oligoadenylate synthetase (2-5 AS) mRNA levels in peripheral blood mononuclear cells (PBMC) of 16 patients with chronic hepatitis B (CHB) and non-A, non-B (CHNANB) during interferon (IFN) treatment. 2-5 ASmRNA was measured by dot blot hybridization using <SUP>32</SUP>Plabeled human 2-5 AScDNA as a probe. 2-5 ASmRNA levels in patients' PBMC, which were similar to those of normal subjects before treatment, increased from 1.2 to 2.7 8h after first dose of IFN and remained high during treatment. 2-5 AS level in PBMC decreased when incubated in vitro even in the presence of IFN, however, 2-5 ASmRNA level increased in response to IFN. This suggets that the decreased 2-5 AS level in vitro may not result from decrease in the 2-5 ASmRNA level, but may be related to events beyond 2-5 AS mRNA production. We also examined the difference in the 2-5 ASmRNA level between patietns receiving different IFN doses. Patients receiving IFN in doses of 1×10<SUP>6</SUP>IU/d had lower level of 2-5 ASmRNA compared to those receiving IFN in doses of more than 3×10<SUP>6</SUP> IU/d.
