When a decoction of a mixture of coptis rhizome and glycyrrhiza root, having a pH value of about 5.5, was left standing, the content of berberine (I) in the decoction gradually decreased. Because I and glycyrrhizin (II) contained in the decoction co-precipitated, and the precipitate was removed from the preparation by filtration. At 20°, the solubility in water of diberberine-monoglycyrrhizinate (III), a main component of the precipitate remained almost constant in the pH range of up to pH 6 and in the pH range above pH 6.5, it increased with pH values. At 80° in all pH range, the solubility of III was generally higher than that at 20°. The solubility of III at pH 5.5 at 80° was 5 times that at 20°. In the presence of II in the pH range of pH 3.5 and 5.5, I was ten times more easily extracted from aqueous phase with 1-octanol, which is due to ion-pair formation.
