To evaluate the chemopreventive activity of motherwort (Leonurus sibiricus L.) on mammary lesions, its effect on precancerous mammary hyperplastic alveolar nodules was examined in female C3H/He retired mice. Free access to drinking water containing a methanol extract of mitherwort at the concentration of 0.5% for 20 days produced in 5-7 and 8-12 month-old mice the nodules which were significantly smaller in both the number and the size than those of the control. There was little difference between the motherwort-treated and the control mice at either age group in any of the pattern of estrous cycle, weights and histological structure of endocrine organs and plasma prolactin level. The results demonstrate that motherwort can inhibit precancerous mammary nodule formation in mice probably with little modulation of the endocrine systems.
