Simultaneous Determination of Main Components in Sho-seiryu-To by Ion-Pair High-Performance Liquid Chromatography

A simple method was established for the precise and simultaneous determination of eight marker components in the Oriental pharmaceutical decoction, "Sho-seiryu-to," and the relevant Kampo extract preparations. The eight marker components, ephedrine, 3,4-dihydroxybenzaldehyde, paeoniflorin, cinnamic acid, methyleugenol, 6-gingerol, schizandrin and glycyrrhizin in samples were separated by high-performance liquid chromatography using an ODS column with a gradient elution system of 10mM tetra-A7-butylammonium phosphate (low UV type, pH4.0)-acetonitrile in which the acetonitrile content was linearly increased from 10 to 80%. The eight components were all eluted within 55 min without being interfered by co-existing components. This method was superior for the simultaneous determination of the components to the other five known methods, i.e, the water-acetonitrile method, the phosphate buffer method, the ion-suppression method, and the ion-pair method using tetra-A7-amylammonium bromide and sodium dodecyl phosphate.
