Antidepressant-Like Effects of Magnolol from Magnolia officinalis in the Forced Swimming Test

The present study investigated the effects of acute magnolol treatment on swim stressinduced alteration of monoamine turnover in the mouse brain. Monoamine turnover was measured in seven brain regions. Forced swimming induced significant decreases in dihydroxyphenylacetic acid (DOPAC) / dopamine (DA) in the striatum and amygdala and 5-hydroxyindoleacetic acid (5-HIAA) / 5-hydroxytriptamine (5-HT) in the amygdala and hypothalamus, and significant increases in DOPAC/DA in the hippocampus, thalamus and hypothalamus and 3-methoxy-4-hydroxyphenylethyleneglycol (MHPG) / norepinephrine (NE) in the amygdala, frontal cortex, hypothalamus, and midbrain. Magnolol (100 mg/kg, i.p.) treatment attenuated swim stress-induced decreases in DA and 5-HT turnover in the amygdala and increases in NE turnover in the amygdala and frontal cortex. Furthermore, administration of haloperidol (0.2 mg/kg, i.p.), a dopamine antagonist, blocked magnololinduced decreases in immobility in the forced swimming test. These results suggest that dopaminergic activity of magnolol in the amygdala may play an important role in antidepressant-like effects.
