We investigated the relationship between gap junction formation and the sex steroids, estradiol and progesterone, in the anterior pituitary gland of ovariectomized female rats. One hundred, 30-day-old female rats of the Wistar-Imamichi strain were prepared and placed into four groups. All rats were ovariectomized on day 30, and they were treated on day 60 with oil, estradiol, progesterone or estradiol and progesterone. Five animals from each treatment group were sacrificed 1, 2, 3, 4 and 5 days after the steroid administation. Both oil and estradiolinjected groups showed no change in gap junction formation. The administration of progesterone to ovariectomized rats increased the number of gap junctions between folliculo-stellate cells, while estradiol injected simultaneously with progesterone blocked the effect of progesterone. These observations provide evidence that the formation of gap junctions within the anterior pituitary glands of female rats is under the influence of the ovarian sex steroid, progesterone.
