Partial Characterization of Elastin Purified from the Skin of the Senescence-Accelerated Mouse

The elastin prepared from the dorsal neck skin of 12〜14-month-old male senescence-accelerated mouse (SAM) were partially characterized. The yield of the elastin from the skin of SAMP1/Yag/Nct (accelerated senescence-prone mice) was significantly higher than that of SAMR1/Yag/Nct (accelerated senescence-resistant mice). The appearance of the elastin of SAMP1/Yag/Nct skin was an amorphous piece, while that of SAMR1/Yag/Nct skin was a fibrous structure. However, the contents of glycine, alanine, and valine which were the major amino acids in the elastin, in that of SAMP1/Yag/Nct skin were not different from those of SAMR1/Yag/Nct skin. Increased elastin in the skin of SAMP1/Yag/Nct would be not changed in its basic structure but its supramolecular one. In the elastin of SAMP1/Yag/Nct skin, the lysine content was slightly lower and the lipid peroxide level was significantly higher than that of SAMR1/Yag/Nct skin. These findings suggested that the elastin of SAMP1/Yag/Nct skin was damaged by oxidative stress. The elastolytic activity of the skin tissue of SAMP1/Yag/Nct was lower than that of SAMR1/Yag. On the other hand, the elastin of SAMP1/Yag/Nct skin showed a very strong resistance on the proteolysis by a porcine pancreas elastase and we thought that this would be one of the main causes for the formation of the elastosis in the skin of SAMP1/Yag/Nct.
