Identification of mutation and homologous chromosomes in four cultivars of Dendranthema grandiflora by physical mapping of 5S and 45S rDNAs using fluorescence in situ hybridization

FISH physical mapping, specific hybridization patterns and the localization of 5S and 45S rDNA genes were investigated simultaneously by fluorescence in situ hybridization (FISH) in the chromosomes of Dendranthema grandiflora (Ramat.) Kitamura cv. 'Shuho-no-chikara'. The FISH detection of 45S rDNA signals was peculiar and revealed 30 signals on 26 chromosomes. Of these, 18 signals on terminal positions of the short arms of 18 chromosomes, eight signals on both the short and long arms of four chromosomes and six signals on the distal region of the long arms of six chromosomes. The detection of 5S rDNA gene revealed eight signals on the long arm of eight chromosomes, of which four signals were found on proximal regions and two signals were found on distal regions. Two chromosomes showed linkage of both rDNA genes, 45S rDNA signal on the short arm and 5S rDNA on the long arm. The mutation of those interstitial signals and both two signals of 45S and 5S rDNA located on the same chromosomes might resulted due to many translocations and/or inversions and recombination, which produced complex and homologous chromosome pairs after long time of breeding this cultivar.
