To understand the mechanisms of formation of hypertrophic scars and keloids, the distribution of various collagens such as type I , III, VI, and XII collagens were immunohistochemically examined in these two abnormal tissues. Types I, III, and VI collagen were positively stained throughout the dermis in early and mature stages of both hypertrophic scar and keloid. Strong expression of type XII collagen was associated with collagen fibers in the early immature phase of hypertrophic scar and keloid. The staining for type XII collagen became faint around thickened collagen bundles and was mostly absent in the nodular structure in mature hypertrophic scar. However, the strong expression of type XII collagen continued to be seen around the thick collagen bundles even after formation of the developed keloids. Our results suggest the presence of distinct molecular mechanisms for formation of hypertrophic scars and keloids. Continuous expression of type XQ collagen throughout the development of keloids may indicate the role of this collagen in progressive formation of collagen bundles.
