Protective Effect of Chronic Administration of 3(2)-tert-Butyl-4-Hydroxyanisole against Spontaneous Mammary Tumors in C3H Mice

In order to examine the effect of 3(2)-tert-Butyl-4-hydroxyanisole (BHA), a preventer of chemical carcinogenesis, on the incidence of spontaneous tumors, it was chronically administered to female C3H, female C3H-BALB-F_1 and male CBA-C3H-F_1 mice by mixing it with a commercial diet. While administration of 0.75% (w/w) BHA in the diet from 6 weeks of age until the end of survival time was associated with a significant decrease in mammary tumor incidence in female C3H mice, average survival time did not change significantly. C3H mice also showed a significant decrease in mammary tumor incidence when 0.375% BHA was administered during the early stages of the life span (from 6 weeks to 6 months of age). When 0.375% BHA was administered from 9 weeks of age to the end of the life span, female C3H-BALB-F_1 mice showed a significant decrease in mammary tumor incidence without any change in average survival time. In contrast to the above observations, administration of 0.375% BHA to male CBA-C3H-F_1 mice from 9 weeks of age to the end of the life span resulted in a slight but significant increase in the incidence of spontaneous liver tumors, and the average survival time decreased considerably. These results indicate that when BHA was administered either during the whole life span or during the early stages of the life span it exerted a protective effect against mammary tumors in female C3H and C3H-BALB-F_1 mice, but not against liver tumors in male CBA-C3H-F_1 mice.
