Studies on the Spectral Shifts and Binding Affinity When Added 4-ene- and 5-ene-Steroids Interact with Adult Pig Testicular P-450 in Androgen Biosynthesis

Spectral shift and binding studies were performed with a purified microsomal cytochrome P-450 from adult pig testis. The binding of substrate and product steroids was measured using spectral methods. There are two pathways in androgen biosynthesis, that is, 4-ene- and 5-ene-pathways. In the 4-ene-pathway, the substrates for progesterone and 17α-hydroxyprogesterone showed typical type I difference spectra. The products such as 4-ene-androstenedione and 5-ene-dehydroepiandrosterone showed reverse type I difference spectra. While, in the 5-ene-pathway, the substrates for pregnenolone and 17α-hydroxypregnenolone also showed typical type I difference spectra. However, 5-ene-substrate steroids exhibited lower affinity than the corresponding 4-ene-steroids.
