α_2-Adrenergic Agonists have no Effect on the Response to Hemorrhagic Shock in Rats

We studied the effects of pretreatment with α_2-adrenergic agonists (clonidine and azepexole) and pentobarbital on hemodynamics during hemorrhagic shock in rats. Hemorrhagic shock was induced by withdrawing blood, and mean arterial pressure (MAP) was maintained at around 40 mmHg. There were no significant differences in survival time, survival rate or heart rate changes between the α_2-adrenergic agonists pretreatment groups and saline treated control group during the hemorrhagic shock in conscious rats. However, in the pentobarbital anesthetized group, survival time was significantly prolonged and survival rate increased during the hemorrhagic shock compared with the control group. Heart rate during the withdrawing period in the pentobarbital anesthetized group was more stable than that in the conscious group. From these results, we had concluded that pentobarbital increased the survival time and survival rate of hemorrhagic shock, however, α_2-adrenergic agonists did not.
