Alteration of Midaglizole Pressor Response with Pentobarbital Anesthesia

To investigate the hemodynamic interaction of α_2-adrenergic drugs and anesthetics, we studied the dose responses of midaglizole (selective α_2-adrenergic antagonist) (0.1-10mg/kg) after administration of saline (1ml/kg), pentobarbital (30mg/kg) or azepexole (selective α_2-adrenergic agonist) (1mg/kg) in rats. Mean arterial pressure (MAP) changes after midaglizole 1, 5 and 10mg/kg were significantly higher in the pentobarbital anesthetized rats than in the saline administrated control rats. MAP after midaglizole 10mg/kg in the azepexole pretreated rats was also significantly higher than the control rats. The combination of pentobarbital and azepexole decreased MAP after midaglizole 0.1, 0.5 and 1mg/kg. These results suggested that the α_2-adrenergic system has an important role in regulating blood pressure during pentobarbital anesthesia.
