Estimation by Pituitary Transplantability of Genetic Resemblance of Three Inbred Strains of Mice

Genetic resemblance was estimated by the pituitary transplantability in 3 inbred strains of mice, C3H/He, GRS/A and SHN, which have no common ancestor(s) retrospectively to at least 50 years ago. All strains of virgin mice were grafted with isologous or heterologous anterior pituitaries, 2 each, under the left kidney capsules at about 50 days of age and plasma prolactin levels at 10 and 20 days after grafting were assayed by homologous radioimmunoassay as an index of pituitary transplantability. The levels of most groups rose over the corresponding control levels at 10 days after grafing. At 20 days, the levels further increased in groups with isologous pituitary grafts. Meanwhile, prolactin levels in mice with heterologous pituitaries were divided into 2 categories; prolactin levels in GRS/A mice receiving pituitaries from SHN changed little between 10 and 20 days after grafting and vice versa. However, GRS/A or SHN mice given C3H/He pituitaries showed marked decline in plasma prolactin levels at 20 days compared to the levels at 10 days and vice versa. These findings strongly suggest that C3H/He mice have the genetic distance greater than that between GRS/A and SHN. No difference between GRS/A and SHN was estimated by the pituitary transplantability in the genetic distance from C3H/He strain. The growth of mammary lobulo-alveolar system was not always parallel to the changes in plasma prolactin levels, the significance of which was discussed.
