A peptide derived from tenascin : C suppresses fibrosarcoma cell growth

【Purpose】 Fibronectin (FN) which is one of the essential extracellular matrix (ECM) proteins, has multiple biological functions : FN participates not only in tissue and organ constructions by providing scaffoldings for cell adhesion, but also in modulation of various cellular processes such as growth, differentiation, migration, and apoptosis through integrin mediated signaling. In these biological processes, integrin - mediated interaction should be controlled reversibly. Based on this standing point, we have demonstrated that FN has a functional site suppressing cell adhesion to ECM. A synthetic FN peptide containing this functional site (termed FNIII14) influences cell growth and differentiation. On the other hand, homology search indicates that there exists several analogous sequences in functional proteins including other ECM proteins and adhesion molecules. Among them, a 22 mer peptide derived from tenascin - C (termed TNIII) was found to induce integrin- mediated cell adhesion to ECM. Here we investigate effects of peptide TNIII on cell growth. 【Methods, Results and Discussion】 Proliferation of normal fibroblast NIH/3T3 in response to PDGF was remarkably stimulated by addition of peptide TNIII in a dose- dependent manner. TNTII also stimulated the proliferation of K-N_78 fibrosarcoma cells (a malignant cell type of NIH/3T3) at a lower concentration (5μg/mL), but suppressed at higher concentrations (>40 μg/mL). To ascertain if TNIII could affect an anchorage independent growth of K-N_78 cells, we tested the effect of TNIII on K-N_78 cell tumorigenesis. When K-N_78 cell suspension (2 × 10^6/200 μL) with various concentrations of TNIII was injected s.c. into the flank of Balb/3T3 mice. In parallel with the in vitro data, TNIII at a low dosage (20 μg/mL) accelerated the tumor growth, but at the high dosage (40 μg/mL) completely inhibited the tumor progression. These results suggest that TNIII may specifically suppress the growth of malignant cell types.
