RECEPTOR-MEDIATED BINDING OF ELASTIN-DERIVED HEXAPEPTIDE TO MACROPHAGES INDUCED A GALACTOSUGAR-SENSITIVE CHEMOTAXIS

Elastin, a highly hydrophobic protein, is a predominant component of mature elastic fibers which confers elasticity and resiliency to the vascular walls, lungs, skin and ligaments. Elastin digests and elastin-derived peptides interact with a variety of cell types to modulate cellular behavior and are implicated to play roles in several diseases such as atherosclerosis, emphysema and metastasis. Our previous study showed polyhexapeptide (VGVAPG)n as a chemoattractant. The circular permutations of VGVAPG were investigated for their chemotactic ability. It was observed that four of the six permutations namely: VGVAPG, GVAPGV, VAPGVG and GVGVAP, were chemoattractant for macrophages and that they serve as signaling peptides which function to regulate macrophage migration. Chemotactic activity induced by these peptides was found to be profoundly sensitive to lactose. Moreover, deactivation study indicated recognition of these four hexapeptides by a single receptor. Macrophages stimulated by VGVAPG demonstrated an enhancement in guanosine 3', 5'-cyclic monophosphate (cGMP) level, thus, reflecting an involvement of cGMP in the signaling mechanism of VGVAPG-induced chemotaxis. However, cGMP elevation was found to be lactose sensitive. It is hypothesized that chemotaxis in response to these elastin peptides is mediated by the action of cell surface receptor. With this working hypothesis. we investigated binding of VGVAPG to macrophages. To facilitate radiolabeling of the peptide for the receptor binding studies, VGVAPG was synthesized with a tyrosine residue to yield Y-VGVAPG. Our report will provide evidence for an elastin-derived peptide receptor present on the cell surface of macrophages.
