INTERACTION OF PROTEOGLYCANS AND α-ELASTIN IN CONSTRUCTION OF EXTRACELLULAR MATRIX OF HUMAN YELLOW LIGAMENT

Human yellow ligament contains abundant proteoglycan (PG) as the matrix factor, and elastic fiber and collagen fiber as the fibrous components, that construct the extracellular matrix. But the interaction between PGs and elastic fiber has not been clarified, therefore we investigated the interaction between PGs and α-elastin from human yellow ligament. One type of large PG and three types of small PGs (decorin, decorin-subtype and biglycan) were purified by ion-exchange chromatography and hydrophobic chromatography, and α-elastin was isolated by alkali treatment from human yellow ligaments taken at the time of operation. The interaction of the PGs with immobilized α-elastin on a sensor was analyzed by surface plasmon resonance. Each of the small PGs had a specific binding to α-elastin, but the large PG had no binding. The binding sites of small PGs were contained in the protein cores rather than in glycosaminoglycan chains. In addition, the differences in the interaction of the small PGs with α-elastin from normal (NYL) and ossified ligaments (OYL) were investigated. The interactions of the small PGs with α-elastin from the OYL were lower than those with α-elastin from the NYL. There were no differences in the amino acid compositions of α-elastin from the NYL and the OYL, but in the OYL, neodesmosine, which is one of the cross-linking amino acids, was significantly less than in the NYL (p<0.05). This finding indicated that there was difference between the OYL and NYL in the interaction of small PGs with α-elastin.
