CHANGES IN EMISSION SPECTRA OF PHOTOSYNTHETIC PIGMENTS IN VIVO

(1) The effects of 3-(4'-chlorophenyl)-1, 1-dimethylurea (CMU) on the fluorescence of photosynthetic pigments in vivo were investigated in blue-green, red and brown algae and in isolated spinach chloroplasts. CMU caused an increase in steady state level of fluorescence of chlorophyll a, but did not influence the fluorescence of phycobilin. The spectrum of the fluorescence increment had a peak at 685 mμ and a shoulder at 730 740 mμ. These two bands probably arise from chlorophyll a (C_<f684>) belonging to pigment system II. (2) On excitation of chlorophyll a in a red alga, Porphyra yezoensis, a fluorescence band with a peak at 720 mμ was observed besides a shoulder at 685 mμ. The 720mλ band is inferred to arise from chlorophyll a (probably, C_<f-1>) in pigment system I. (3) On addition of CMU to the algal cells, the induction of fluorescence was modified to take a simple time course. The Induction was observed only with respect to the fluorescence of chlorophyll a, but not in the fluorescence of phycobilins. The spectrum of the "transient" fluorescence showed two emission bands of chlorophyll a at 685 mμ and 740 mμ and was quite similar m form to the srectrum of the CMU-caused increase in steady state fluorescence. (4) These facts were interpreted In terms of the correlation of fluorescence of chlorophyll a and the photochemical reactions of photo-synthesis
