Preparation of girdle lamella-containing chloroplasts from the diatom Phaeodactylum tricornutum

Chloroplasts were isolated from the diatom Phaeodactylum tricornutum by French press treatment and centrifugation. Electron micrographs of the isolated chloroplasts indicated that they lacked most of the envelope membranes but retained the lamellar structure characteristic of the diatom chloroplast; three thylakoids were grouped to form a band which transversed the chloroplast. A girdle lamella also composed of three thylakoids surrounded these transversal lamellae. The isolated chloroplasts were active in photosynthetic electron transport reactions including the Hill reaction, the Mehler reaction and the system I reaction.
