Specific Interaction of Nuclear Protein HBP-1 with the Conserved Hexameric Sequence ACGTCA in the Regulatory Region of Wheat Histone Genes

The plant histone genes sequenced so far have conserved hexameric (ACGTCA) and octameric (CGCGGATC) sequences in their 5'-flanking regions. We proviously showed that the nuclear protein HBP-1 binds to the hexameric sequence of wheat histone H3 and H4 genes. Here we report several lines of evidence that confirm the existance of this DNA-binding protein and describe the results of our analysis of its binding properties. A DNase I footprinting experiment demonstrated that HBP-1 covers a sequence consisting of approximately 20 bp in which the hexameric motif is located at the center. A mobility shift assay with synthetic oligomers used as the binding competitor showed that HBP-1 strictly recognized its binding site. HBP-1 could not bind to the core sequence (CACGTGAC) that is essential for the binding of major late transcription factor (MLTF) even through there was high homology between the target sequence (ACGTCA) of HBP-1 and the MLTF binding sequence. The core sequence for the binding of activating transcription factor (ATF) was identical to that for HBP-1 binding. We concluded that HBP-1 is a transcription factor for plant genes that is analogous to ATF.
