Specific Binding of Nuclear Protein from Tobacco Hairy Roots Cultured in Vitro to a 5'-Upstream Region of the rolC Gene of the Ri Plasmid

The site of initiation of transcription of the rolC gene (ORF12) in the TL-DNA of the Ri plasmid was determined by the protection analysis with S1 nuclease. The initiation site was located 27 bp upstream from the first ATG codon of the rolC gene. A CAAT box and a TATAA box were found at -57 bp and -35 bp, respectively. The specific binding of a nuclear protein isolated from tobacco hairy roots cultured in vitro to the 5' non-coding region of the rolC gene was detected by gel-retardation analysis. Footprinting with DNase I revealed the apparent protection of a region from -83 bp to -60 bp where a sequence identical to the AT-1 box (AATATTTTTATT) of the promoter of light-inducible genes (Datta and Cashmore 1989) was found.
