Requirement of Manganese for Electron Donation of Hydrogen Peroxide in Photosystem II Reaction Center Complex

Mn^<2+> was required for the electron donating reaction from H_2O_2, but not for that from diphenylcarbazide (DPC), in the PS II reaction center complex which was prepared from spinach chloroplasts by Triton X-100 extraction. The reaction center complex showed a high activity of 2,6-dichloroindophenol (DCIP) photoreduction in the presence of DPC, but a low activity with H_2O_2. The H_2O_2-supported DCIP photoreduction was suppressed by EDTA and enhanced by a small amount of Mn^<2+>. Ca^<2+> and Mg^<2+> could not replace Mn^<2+>. The activation by Mn^<2+> and its binding showed two binding sites of Mn^<2+> in the reaction center complex, with high (1.5 × 10^7 _M^<-1>) and low (1 × 10^6 _M^<-1>) binding constants.
