Carotenoid photobleaching induced by photosystem II action was studied using membrane fragments of the blue-green alga Anabaena variabilis. Special attention was paid to the action of O_2. Carotenoid photobleaching elicited by carbonyl cyanide m-chlorophenylhydrazone (CCCP) depended on O_2. However, the addition of H_2O_2, sodium silicotungstate or potassium ferricyanide (Ferri) , an electron acceptor for reaction center II action, removed the O_2-dependency. These results indicate that O_2 acts as the electron acceptor for this reaction. When both CCCP and Ferri were present, a short illumination (0.25 sec) caused a rapid photobleaching followed by a slow recovery in the subsequent dark period. The spectrum of the absorption decrease in the light was identical with that of the absorption increase in the subsequent dark, indicating that a reversible process is involved in the carotenoid photobleaching. The size in the dark recovery relative to the light bleaching became larger under anaerobic conditions and smaller under higher partial pressure of O_2. The reuslts were interpreted as indicating that O_2 does not function in the primary process including a reversible bleaching step, but is involved in the slow and irreversible bleaching process.
