Regeneration of photosystem II and phycobilin pigments in photoorganotrophically grown Anabaena variabilis

Regeneration of photosynthetic activity and phycobilin pigments was studied with cells of Anabaena variabilis lacking photosystem II activity and phycobilin pigments. Regeneration was achieved only when the cells were incubated in the presence of nitrate or nitrite. The addition of ammonium salts or urea was far less effective. Nitratc-directed regeneration was independent of light and inhibited by chlorate. Dark-regenerated cells, however, differed from light-regenerated ones in that the former were incapable of excitation transfer from phycocyanin to pigment system II chlorophyll a, although they emitted fluorescence of pigment system II chlorophyll a origin, if illuminated by the light absorbed by chlorophyll. The regeneration process in Anabaena cells is assumed to consist of two steps: [1] light independent, nitrate-supported synthesis of phycobilin pigments and photosystem II integrity, followed by [2] light-directed formation of excitation transfer from phycocyanin to pigment system II chlorophyll a. An antibiotic study revealed that the former is associated with protein synthesis, while the latter is not.
