Intracellular ATP level and light-induced inhibition of respiration in a blue-green alga, Anabaena variabilis

To evaluate the possibility that the respiration of photosynthetic organisms competes with photophosphorylation for ADP in vivo, we determined the intracellular ATP level in cells of Anabaena variabilis under various respiratory conditions; maintained in darkness, light-inhibited or uncoupled. No substantial difference in ATP level was observed between the light-inhibited and dark-kept cells. Under dark anaerobic, conditions, however, the ATP level decreased to nearly one-third that of the aerobic level, indicating a capacity for oxidative phosphorylation in this alga. In the presence of 10^<-5> M CCCP, the ATP level was reduced to about half that of the control, but the light-inhibited respiration was not eliminated by this agent, Suggesting that it is not due to the decreased ADP level. A possible mechanism for the photoinhibition is discussed with special reference to the procaryotic intracellular structure of this alga.
