In vivo chlorophyll forms in higher plants and algae sensitive to treatment with lutein

Lamella preparations of spinach, Chlorella, Phaeodactylum, Anabaena and Porphyra were treated with a hydrophobic reagent, lutein, and the absorption and fluorescence spectra in the red region before and after treatment were compared for changes caused by the treatment. Absorption spectra of all these preparations underwent the same spectral change, transformation of a band at 684 nm into a band at 666 nm. The longer the maximum wavelength of the red peak, the greater was the fractional absorbance decrease at 684 nm. The content of C684 (the chlorophyll form responsible for the 684 nm band) in the lamellae was estimated from the fractional decreases as being progressively higher in the order of Phaeodactylum. Porphyra. Anabaena. Chlorella and spinach. The fluorescence spectra at liquid nitrogen temperature before treatment showed two bands. The longer wavelength band was transformed by the treatment into a shorter wavelength band(s), as described below, according to the maximum wavelengths: spinach, F735→F695 (or F686); Chlorella, F715→F700 (or F686) ; Phaeodactylum, an unidentified component→F690; Anabaena, F732→F685 (or F695); Porphyra, F726→F683. These chlorophyll forms with fluorescence maxima between 715 and 735 nm were, therefore, designated C684 based on absorption spectrophotometry, and are considered to play a role in photosystem II.
