Two new methods were developed for the determination of swertiamarin, the main bitter principle of the herb of Swertia japonica and of Swertia pseudochinensis. In the preparative thin-layer chromatography-ultraviolet (TLC-UV) method, swertiamarin in the methanol extractive of Swertia herb is separated by preparative TLC developing with AcOEt・n-PrOH・H_2O (6 : 1 : 3) (detected as a pink spot by a PAN-UV Lamp) and is determined by the ultraviolet absorbance at 238nm. In the TLC-densitometer (DM) method, the MeOH extractive containing swertiamarin is developed on a TLC plate (Kieselgel 60F_254) using the same solvent system, and the amount of swertiamarin (Rf ca. 0.3) is determined by TLC densitometry, using a dual-wavelength TLC scanner. By these methods, the contents of swertiamarin were found to be 1-2.5% in Swertia Herba and 0.9-1.5% in Swertia Pseudochinensis Herba (both Osaka market products). It has also been found that the content of swertiamarin in Swertiae Herba varie depending on the parts of the original plant and decreases in the order of flowers, leaves, stems and roots.
