Cell proliferation kinetics were studied on the matrix cells of the chick embryo telencephalon by means of a ^3H-thymidine autoradiography. The mitoses per unit length of the lower most matrix layer were most abundant at 6 days of incubation (6-D) and decreased thereafter, being considerably fewer at 10 days (10-D). Therefore 6-D and 10-D were selected as two time points representative of high and low proliferation activities. An injection of ^3H-thymidine into an egg resulted in a gradual increase of labeling index with time, indicating a continuous labeling of DNA. The growth fraction was obtaind as a plateau of labeling index and decreased from 0.89 at 6-D to 0.52 at 10-D. The cell cycle phases were calculated from this lateling index curve and from the percent labeled mitoses curve. The generation time increased from 19.8 hr at 6-D to 23.6 hr at 10-D. The duration of G_1, S, G_2 and M phases were 7.9, 8.6, 2.7 and 0.6 hr at 6-D and 7.0, 13.6, 2.0 and 1.0 hr at 10-D, respectively. We conclude that prolongation of T_c was caused mainly by prolongation of T_s.
