The effects of ultraviolet-B (UVB) irradiation on the skin : Induced apoptosis and Photoreactivation

Potential causes for the global decline of amphibians include environmental changes. The diminishing ozone layer and consequent increase of ultraviolet-B radiation reaching the earth's surface has been hypothesized to be a modality factor. A deleterious change in DNA, especially the production of pyrimidine dimers is a main effect of UVB exposure. UVB-irradiation-induced apoptosis in the dorsal skins of anuran juvenile Bombina orientalis was investigated by the new TUNEL method and electron microscopy. After UVB irradiation, many TUNEL-positive cells were found in the epidermis. Typical apoptotic figures were observed in the cell fragmentation and nuclear chromatin condensation in the periphery of the nuclei. (Sasaki & Abe: Electron microscopy vol. 36 supple 1 234 2001) Also, we tested the relationship of productivity to DNA damage and repair. Electrophoresis image analyzing for samples of UVB- and/or blue light-irradiated skin was carried out to obtain measurements for photoreactivation and excision repair. Photoreactivation and excision repair was recognized in the skin irradiated by UVB (3,8 KJ/m^2). Therefore, it can be said that the photoreactivation system is in this Bombina orientalis.
