Expression of the bile salt export pump and multidrug resistance protein 2 in liver development

We examined the gene and protein expression as well as distribution of antigenic sites of Bsep and Mrp2 in the rat liver during development from 16-day-old fetus to adult animal. Bsep mRNA was almost undetectable before, but clearly expressed after birth. In contrast, Mrp2 mRNA was seen before and after birth. Expression of Bsep and Mrp2 proteins corresponded to the expression of their mRNA. Immunofluorescent microscopy showed asynchronous appearance of Bsep and Mrp2 during development, but their colocalization at the canaliculi once each one is expressed, thus indicating early polarization of these canalicular transport proteins. These findings provide molecular evidence of the transcriptional regulation of the expression of Bsep and Mrp2 during development and indicate the different relevance to the liver function of the systems responsible for canalicular trasport of bile salts versus organic anions.
