The bactericidal effect of unsterilized night soil against colon bacteria and some intestinal pathogens at mild temperature was demonstrated by the following convenient method for anaerobic culture. Inoculated medium is sealed in a small pouch made of polypropylene film, dipped in 3% Na_2SO_3 awueous solution, which absrobs dissolved oxygen through the film, and after incubation, viable cells are counted on selective media. Escherichia coli K-12,Salmonella typhimurium and Vibrio parahae molyticus suspended in unsterilized night soil barely survived more than 3 days at 20℃, and died off at 50℃ within 1 day of incubation under anaerobic condition. E. coli died off within 1 day at 30℃ and 40℃, S. typhimurium died off within 2 days at the same temperatures, and V. parahaemolyticus died off within 1 day and 2 days of incubation at 30℃ and 40℃, respectively, while all strains grew well over 3 days at 30℃ and 40℃ when night soil was replaced by nutrient broth.The bactericidal effect of night soil against intestinal bacteria may be partially explained by the generation of ammonia from urine, as indicated by the following experiment. Human feces were diluted 10 times with tap water or human urine, inoculated with about 5×10^9/ml each of E. coli and S. typhimurium, and incubated anaerobically at 30℃ for 40 hr. Viable counts of these microorganisms decreased to about 1/12 and 1/5 in the case of dilution with water, but to about 1/4500 and 1/1400 in the case of dilution with urine.
